What percentage of peptides actually works?
There is no single 'peptide percentage' — the studied doses span three orders of magnitude depending on which peptide you mean. Palmitoyl pentapeptide-4 (Matrixyl) has RCT evidence at 3 parts per million; argireline's originating study used 10%. Palmitoyl pentapeptide-4 has the most credible independent human trial data of any cosmetic signal peptide, and the studied concentration is startlingly low. A 12-week, double-blind, vehicle-controlled, split-face RCT in 93 women aged 35-55 tested a moisturizer containing pal-KTTKS at 3 parts per million (0.0003%) and found statistically significant improvement in wrinkle and fine-line scores versus vehicle, confirmed by both image analysis and expert grading. The honest caveat cuts the other way from most 'higher % is stronger' assumptions: at ~802 Daltons, pal-KTTKS is well above the molecular-weight threshold generally associated with passive skin penetration, and permeation studies found neither KTTKS nor pal-KTTKS detectable in the receptor compartment of full-thickness skin models. Matrixyl 3000 (palmitoyl tripeptide-1 + palmitoyl tetrapeptide-7) and Matrixyl Synthe'6 (palmitoyl tripeptide-38) are newer signal-peptide complexes marketed as upgrades on the original Matrixyl. The CIR Expert Panel's safety review of tripeptide-1, hexapeptide-12, their metal salts and fatty acyl derivatives, and palmitoyl tetrapeptide-7 notes typical cosmetic use concentrations below 10 parts per million.
